Antibacterial Activity of Ethanol Extract, n-Hexana, Ethyl Acetate And Water Fraction From Wangon Leaf (Olax scandens Roxb.) Against Staphylococcus aureus ATCC 25923
Abstract
Wangon leaves contained chemical compounds such as saponins, tannins, flavonoids,
steroids and phenols. The aim of this study was to determine antibacterial activity of ethanol
extract, n-hexane, ethyl acetate, and water fraction from wangon leaves to Staphylococcus
aureus ATCC 25923 bacteria.
Wangon leaves powder was extracted by maceration method with solvent ethanol
96%. Wangon leaf extract that has been obtained then was fractionated using n-hexane, ethyl
acetate and water as solvent. Ethanol extract and fractions were tested for its antibacterial
activity using a diffusion test with 50%, 25%, and 12.5% concentration to find out which
fraction was the most effective. The most effective fraction was tested in dilutions method
with various concentrations ie, 50%; 25%; 12.5%; 6.25%; 3.125%; 1.562%; 0.781%;
0.391%; 0.195%; 0.098% to determine the minimum inhibitory concentration and minimum
kill concentration.
The results showed that the ethanol extract, n-hexane, ethyl acetate, and water
fraction had antibacterial activity against Staphylococcus aureus ATCC 25923. The 50%
concentration of ethyl acetate fraction was the most effective against Staphylococcus aureus
ATCC 25923 and had an inhibition zone diameter of 12.5 mm. But the Minimum Inhibitory
Concentration was unknown and Minimum Kill Concentration was 25%.
